Abstract (EN)

In this thesis study, first of all, after the reaction of 2-chloro benzohydrazide and hexyl isothiocyanate reagents in Tetrahydrofuran solvent, 5-(2-chlorophenyl)-4-hexyl-4H-1,2,4-triazole-3-thiol compound was obtained in basic medium. Afterwards, derivatives of this compound were synthesized using different fatty acids such as Louroyl Chloride, Myristoyl Chloride, Palmitoyl Chloride and Stearoyl Chloride by removing the acidic proton. Then, the structures of the obtained compounds were characterized using 1H-NMR, 13C-NMR and FT-IR. Optimized three-dimensional structures, HOMO-LUMO energy levels and MEP map were calculated theoretically by DFT techniques using Gaussian 09 program. MTT method was used to determine the antitumor activity of compounds synthesized in HT-29 and HCT-116 colon cancer cells and MDA-MB-231 triple negative breast cancer cells. By examining the antitumor properties of the synthesized compounds with the MTT method, the determination and comparison of the biological activities as a whole was based on the studies.
