Molecular-physiological detection as a more accurate diagnostic method to detect some diarrheal parasitic infections in Al-Muthanna province

Abstract (EN)
Diarrhea is one of the most common health problems, there are many possible causes of chronic diarrhea, such as that causes by microorganisms as Parasites Bacteria, and Viruses, however, some causes of chronic diarrhea sometimes remain unknown. The aim of our study is detection of the co-infection of some intestinal parasites diarrheal patients. The current study was carried out in Al-Muthanna Province which included collection of stool samples from diarrheic patients who were consulted in female and children's Hospital of General Al- Rumatha Hospital at a period extended from Febraury / 2021 – June / 2021. 100 people stool samples taken with different ages to both sexes and examined by light microscope for detection of intestinal parasites, the results showed that the percentage of positive samples with INPs are (78.0%), Negative (22.0%) other causes of diarrhea, E. histolytica was predominant parasite highest percentage ,followed by G. lamblia , while the lowest rate with C. parvum infection. There are non-statistically significant variations in infection rates by gender, residence, and age groups; nonetheless, men are infected at a higher rate than females. The most people in rural areas were infected than in urban areas.The age group with the greatest infection rate (less than 1 – 10 years) as well as the youngest age group (11-20 years). A total of 100 microscopy examination samples are examined by PCR for diagnosis of E. histolytica, G. lamblia, and C. parvum, , (96%) positive samples with intestinal parasites, and (4%) negative is the dominant parasite E. histolytica with (53.8%), followed by G. lambliawith (34.4%) and the lowest parasitic infection with (11.8%) C. Parvum infected There are non-statistically significant differences according to gender habitation, and age groups . However, the most infection was with males more than females and the most infection was in rural more than an urban population, and the highest infection rate in age group (less than 1 – 10 years) and the lowest age group (11-20 years). The DNA sequencing method was carried out to genetic confirmative detection and genetic variation (substitution Mutations) analysis for small subunit ribosomal RNA genes in local Entamoeba histolytica, Giardia intestinalis, Cryptosporidium parvum IQS isolates that compared with NCBI-Blast related isolates . Finally, the local Entamoeba histolytica, Giardia intestinalis, Cryptosporidium parvum IQS isolates were submitted into NCBI Genbank and identified by accession numbers (OL719307, OL719308, OL740025).
