Determination of polyphenolic compounds in plant product matrices by high performance liquid chromatografy method

Abstract (EN)
In this study, high performance liquid chromatography (HPLC) method has been developed and validated for determination of important polyphenolic compounds including ellagic acid (EA) and rutin (RUT) in the plant product matrices. EA and RUT were determined in sambucol (black elderberry) samples which used as food supplements, some molasses and pomegranate juice by using photodiode array detector (DAD) at 254 nm wavelength value within 18 minutes on a phenyl column (4.6 mm×100 mm×3 µm). A (water:methanol:formic acid; 88:10:2; v/v/v) and B (water:methanol:formic acid; 8:90:2; v/v/v) solutions were used as mobile phase. The flow rate was 1.0 mL/min and the injection volume was 20 µL, respectively. In the experiments two internal standard (IS) substances were used including methyl paraben and ethyl paraben. Matrix-matched calibration curve method was applied in the experiments. Linear graphs were obtained with high correlation coefficients (r2= 0.9916-0.9985) for EA and RUT. In intra-day and inter-day precision studies with less than 7.4% RSD% values were obtained for EA and RUT. The developed method has been successfully applied to the plant product matrices.
