Abstract (EN)

Cancer, which is one of the most common diseases today, has been the subject of many studies. Due to its high mortality rate, this subject has been studied frequently and various treatment methods have been investigated. Cancer neither can be treated with a single method nor can it be treated with a single drug. Therefore, many studies have been carried out on various drugs. This study aims to investigate the effects of Metformin, one of the important discoveries of recent years, and Cisplatin drugs, which are used in cancer treatment, on the cancer cell line. According to the results of the toxicity analysis, Metformin LD50 value: 43 mM, Cisplatin LD50 value: 23 μM. Over these values, the multiples of Metformin and Cisplatin drugs were studied individually and in combination on the HeLa cell line. HeLa cell lines have been used by raising as 3D and 2D cell models. 3D cell models have been preferred because they are an alternative to in vivo studies. In this study, the drug doses applied to 2D cell lines do not show their effect in the 3D model structure. Similar to the systemic structure, it has been concluded that higher doses are needed for the death of cancer cells. In the light of studies on 2D and 3D model structures, it has been observed that Metformin is effective on cervical cancer and may be a potential drug. The fact that the effect of metformin is more dominant when given in combination with cisplatin also shows the efficacy of this drug.
