Abstract (EN)
Propagation of Hartwissian Oak (Quercus hartwissiana L.) Using by Tissue Culture Techniques Micropropagation techniques are very important methods for clonal propagation of selected forest trees. In this study, producing of hartwissiana oak {Quercus hartwissiana) with axillary bud culture was aimed. Explants were cultured in GD and WPM for ten weeks until the shoot multiplication was observed. Different doses of BA were applied to each nutrient medium. The best nutrient medium was GD medium in respect to shoot number, nod number and shoot length The best doses of BA were found to be 1 mg/1 for initial culture and 0,2 mg/1 for shoot multiplication. The explants belonging to 7 different genotypes were- cultured into GD medium supplement with 0,2 mg/1 BA. Shoot and the nod number even not different but differences were observed in shoot length depending on the genotype. Influence of explants source were different on shoot development and shoot multiplication. Explants from apical bud have the lowest multiplication rate. However Axillary bud was more effective in shoot number, nod number and shoot length The two separate experiment were carried out for the propos of rooting. In the first Microshoots placed in 1/3 stranght GA medium after their base had been dipped in 0,5 -1 mg/1 BA for 2-6 minutes. 35 days later rooting rate were found as 13 %. In the second experiment, shoots were keep in nutrient medium containing 25 mg/1 BA for 24 and 48 hours and subsequently transfer to auxin-free medium containing % 1 activate charcoal. As a result the best rooting was observed as 100 % and 80 % for 48 hour and 24 hour treatment in media with BA respectively.
