Abstract (EN)

This thesis explores the development of novel polymeric materials to address challenges in disease diagnosis and drug delivery. This thesis's first and second studies are related to developing novel polymer-based drug delivery systems. The first study synthesized different polymer-drug conjugates to find the best candidate for in vitro performance towards pancreas adenocarcinoma. Drugs were conjugated to the polymer backbone via pH and/or redox-responsive linkers, and drug release profiles and cytotoxic activities against different cancer cells were evaluated for each copolymer. In the second study, biodegradable nanogels for targeted drug delivery were fabricated using self-assembly, and the drug was physically loaded into nanogels to achieve efficient drug delivery vehicles. In the third part, biodegradable and injectable hydrogels were fabricated using a metal-free click reaction. Azide and gemcitabine-containing copolymers containing carbamate and disulfide linkers between the drug and polymer backbone were synthesized and crosslinked using cyclooctyne-terminated telechelic poly(ethylene glycol) polymer. Hydrogels were evaluated for drug release and cytotoxic activity. The other projects reported in this thesis focus on the fabrication of novel polymeric coatings for biomedical applications such as biomolecular immobilization, cellular attachment, and release. In the fourth study, hydrophilic polymer brushes were prepared and functionalized using a radical-exchange cross-coupling reaction to develop sensing and protein immobilization platforms. In the fifth project, thiol-reactive redox-responsive polymer brushes containing pyridyl disulfide units were fabricated, and functionalization with thiol-containing dyes, drugs, and biomolecules was shown. In the sixth study, thiol-reactive benzothiazole-disulfide-containing polymeric thin films were prepared and investigated for cellular attachment and cell release studies. In the final study, adamantane-containing polymeric thin films were fabricated, which could undergo supramolecular interaction with functional ligands bearing beta-cyclodextrin, and modified surfaces were used for protein and cell attachment.
