Abstract (EN)

Many molecular genetic methods and new technologies which are based on DNA base, have been used for determining identity, criminals in killing and physical abuse and parents in forensic medicine. The aim of this study is to evaluate legal use of molecular methods which are used, commonly. DNA isolation had been made in pursuance of isolation protocols of manuel and M48 biorobot from various regions of Turkey the forensic cases which come to Biology Expert Bureau of İstanbul Forensic Medicine Foundation Management. However, evaluations had been made in respect of the results of otomatic isolation. Obtained DNA isolates had been reproduced or increased in respect of touch down PCR protocol by using isolation kit by Qigen mark. Typecasting had been made by capiller elektroforez method in ABI Prism 3130x tool. Evaluation had been made or applicated by using the methods of RFLP (Restriction Fragment Lentht Polymorphism), PCR 8Polymerase Chain Reaction), VNTR (Variable Number Tandem of Repeat Polymorphism), ASO (Allel Specific Oligonucleotid Dot Blot Analyses), STR (Short Tandem Repeat Polymorphism), X-STR, Y-STR according as demanded analyses. In this study, materials which are scene of crime, with capiller elektroforez views or images overlap as one to one in many of the events which were occured. It is argued that PCR, STR, X-STR, Y-STR methods are more used in relation to the other methods in identification applications in forensic medicine.Key Words: Molecular Genetic Methods, PCR, RFLP, VNTR, ASO, STR, X-STR, Y-STR
