Trans-activation of AMV197 belong to amsacta moorei and its role in the host range of virus

Abstract (EN)

Previous researches suggests that Amsacta moorei entomopoxvirus open reading frame AMV197 encodes a serine/threonine protein kinase, and a recombinant virus (Am-pk/gfp) was created by changing this gene with green fluorescent protein. Based on these studies it is thought that pk plays an important role in virus replication and gene regulation. In order to understand the the role of pk gene in host specificity and virus proteins in the virus replication, I have studied the transactivators of AMV197 and compared wild type and recombinant virüs replication in various cell lines. In order to determine proteins that has role in the transactivation of pk, South-western hybridization assay was performed. This study showed that cell proteins (50, 38, 35, 27 ve 16 kDa) play role in the transactivation of pk. In order to determine the role of pk in virus replication, wild type and recombinant virus were assayed in different cell lines (Lymantria dispar, Ld652 (control); Bombyx mori, Bm; Spodoptera frugiperda, Sf9) for virus yield and DNA replication. Slot-Blot Hybridization and End Point Dilution Assay (EPDA) showed that host factores play importans role during virus replication.
