Investigation of prokaryotic diversity of some thermal springs in Balikesir and Kutahya district

Abstract (EN)

In this study, prokaryotic diversity of Balıkesir-Sındırgı, Gure, Havran and Kütahya-Eynal thermal sources were investigated by culture-dependent and culture independent methods. After the incubation 66 strains were isolated and 16S rRNA amplified with specific primers and then ARDRA analysis of amplification products were performed by cutting with the restriction enzyme Mbo I. Result of sequencing, isolates which belong to the genus of the Geobacillus, Bacillus, Aeribacillus, Anoxybacillus, Brevibacillus, Chelatococcus and Aneurinibacillus were obtained. In addition, the FAME analysis was applied to the some isolates having different ARDRA profiles. 16S clone library was created within the scope of culture-independent methods and FISH studies were conducted. As a result of cloning research, we have obtained 20 different ARDRA profile and after sequencing clones belonging to the genus Bacillus, Geobacillus, Anoxybacillus, Meiothermus, Rosemonas, Aneurinibacillus and Brevibacillus were obtained. As a result of FISH study, members of the domain Bacteria signals have been received, but members of the domain Archaea signals have not received.
