Investigation of genomic DNA of some of the Tabanus (Tabanidae, Diptera) specieses by RAPD-PCR technique

Abstract (EN)

In systematic studies, many species have been classified according to morphological criteria. However, recently occuring environmental pollution and climatological changes leads to some changes in morphologic features of organisms. Together with morphologic properties, using molecular markers for identification of species, determination of genetic distance and detecting polymorphism is quite important. Randomly Amplified Polymorphic DNA Polymerase Chain Reaction (RAPD-PCR) technique is preferred in order to determine genetic distances since it is economical and rapid and also it does not require genomic information.In this study, the amplification of 20 primers with genomic DNA isolated from some Tabanus (Tabanidae Diptera) species is tested. According to the amplification results of 3 primers tested, the degrees of diversity from one another are showed. It is demonstrated that RAPD-PCR technique could be used to determine genetic distances for Tabanus species.Keywords : Tabanus, RAPD-PCR, Genetic distance, Phylogeny, DNA
