Investigation of the possible protective effect of chlorogenic acid in rats subjected to oxidative stress inducted by 2,4- D (Dichlorophenoxy acetic acid), a herbicide

Abstract (EN)

The aim of this thesis study is to determine the curative effect of Chlorogenic acid (Chloro) on oxidative stress, histopathological and immunohistochemistry changes induced in blood, liver, heart, kidney, brain and ovary tissues by 2,4 D (Dichlorophenoxy Acetic Acid) based herbicide causing subchronic toxicity. Approval was obtained from Pamukkale University Animal Experiments Ethics Committee for the application with the number E-60758568-020-255262. In the study, a total of 28 Wistar male rats (300-350 g, 10-12 weeks old) were divided into 4 groups by random sampling method with 7 animals in each cage. Group I (control group) was given normal rodent feed and tap water ad libitum during the study and saline was administered orally. Chlorogenic acid 10 mg/kg was given to Group II (KLORO), 2,4 D (Dichlorophenoxy Acetic Acid) based herbicide 50 mg/kg was given to Group III (2,4-D) and 2,4 D (Dichlorophenoxy Acetic Acid) based herbicide 50 mg/kg and chlorogenic acid 10 mg/kg were given to Group IV (2,4-D+KLORO) by oral gavage once a day for 28 days. 2,4 D (Dichlorophenoxy Acetic Acid) significantly increased MDA levels in blood, liver, heart, kidney, brain and ovarian tissues, while it decreased GSH levels in these tissues and SOD and CAT activities in tissues and erythrocytes. In contrast, these parameters were found to improve in the KLORO-treated group. In addition, KLORO corrected the histopathological changes induced by 2,4 D (Dichlorophenoxy Acetic Acid) in rat tissues. In conclusion, KLORO prevented oxidative stress by inhibiting lipid peroxidation induced by 2,4 D and activating the antioxidant defence system and improved the damage caused by 2,4 D in blood, liver, heart, kidney, brain and ovarian tissues of rats.
