EFFECTS OF EVEROLIMUS ON OXİDANT-ANTIOXIDANT SYSTEM AND ONRENAL HISTOLOGY IN EXPERIMENTAL RENAL-ISCHEMIA REPERFUSIONINJURYAcute Renal Failure is frequently seen in clinical settings and the major cause of acute renalfailure is ischemia-reperfusion injury. Therefore, it is important to explain the pathogenesis ofischemia-reperfusion injury and to find a prophylactic treatment of the prevention.In this study 46 adult Wistar albino male rats were seperated into three groups. Group I wasthe control group and no treatment was given to the rats in this group. Blood samples weretaken and kidneys were harvested after laparotomy. Group II was ischemia-reperfusion groupand no treatment was given to the rats in this group. Group III was the treatment group andrats were given everolimus by gastric gavage for 7 days in a dose of 1.5 mg/kg/day in thisgroup. Group II and group III rats underwent 45 minutes of bilateral renal ischemia and ineach group following 2 and 24 hours of reperfusion blood samples were taken and kidneyswere harvested.Blood samples were taken for analysis of serum BUN, creatinin, IL-6 and TNF-α levels andkidneys were harvested for tissue MDA and MPO levels, SOD activity, and histopathologicalexamination. The results indicate that, although everolimus improves oxidative stres,cytokines and inflammatory markers in ischemic renal injury, it does not seem to be adequatein the prevention of renal functions and improvement of renal histology, because of itsprobable toxicity to the tubular cells.
