Comparison of the effects of desflurane and sefovlurane anesthesis odministered on hepatic tissues in rats with different age and gender

Abstract (EN)
The purpose of this study was to determine both the hepatic effects of desflurane and sevoflurane, and the importance of age and gender on toxic effects. Following local research ethics committee approval, 81 rats were first divided into 4 groups: 21 young male rats, 21 young female rats, 21 old male rats and 21 old female rats. Then, each group was further divided into 3 groups: control, desflurane and sevoflurane groups. Mamtaining minimum alveolar concentration (MAC) 1 for rats, 6% desflurane and 2% sevoflurane were applied in 6 L of 100% oxygen for 2 hours in a 40x40x70 cm bell glass. At the end of anesthesia, all rats received ketamin 3 mg kg-1 intraperitoneally and euthanasia was applied by taking intraabdominal blood. Hepatic capsules of rats were dissected and put into 10% formaline solution and sent to pathology laboratory for hystopathological examination. After routine fixation, liver tissues were taken into parafine blocks and 5(x sections were stained with hematoxylin and eosin. Hystopathological examination was done by a light microscope. Hydropic degeneration (HD), nuclear polymorfism (NP), portal neutrofil infiltration (PNI), portal lenfosit infiltration (PLI) and focal necrosis (FN) were determined in each preparate and injury criteria was evaluated. Each preparate was scored between 0 and 3 and hystopathological injury fact score (HIFS) and total injury score (TIS) were obtained and mean injury score (MIS) for each group was calculated. In conclusion, desflurane and sevoflurane had no effect on young male rats, but mild- moderate hepatic injury was found in old male-female rats and young female rats, predominantly in old rats. Hepatic injury in old female rats was more with desflurane compared to sevoflurane. As hepatic injury due to these two agents is mild-moderate, we believe that they may be safely administered in every day anesthesia practice.
