Investigation of the protective effect of cinnamaldehyde against doxorubicin-mediated nephrotoxicity

Abstract (EN)

Doxorubicin (DXR), which is widely used in the treatment of cancer, one of the most important health problems of today, is used in the treatment of many cancers such as hematologic, solid and soft tissue sarcomas. One of the main factors limiting the therapeutic use of DXR besides its cancer cell destroying effect is that it causes nephrotoxicity. It is necessary to identify agents that will reduce the damage caused by DXR without altering its role in treatment. In this study, it was aimed to investigate the protective and ameliorative effects of Cinnamadehyde (CA) on kidney tissue on DXR-induced nephrotoxicity. In the study, 48 rats divided into 6 groups as Control, CA10, CA20, DXR, CA10 + DXR and CA20 + DXR were used. Cumulative administration of 6 doses of DXR (21 mg/kg, intraperitoneal) increased serum BUN, creatinine and uric acid levels as well as renal lipid peroxidation and decreased GSH levels and antioxidant enzyme activities of SOD, CAT and GPx. DXR also increased NF-κB, TNF-α, IL-1β, iNOS levels and p38α MAPK activity and decreased IL-10 levels. KIM-1 and NGAL levels, which are markers of acute kidney injury, were also increased by DXR administration, while AQP 1 level was decreased. In addition, DXR activated apoptosis by increasing apoptotic Bax and Cyt-c expression levels and decreasing antiapoptotic Bcl-2 and Procas-3 expression levels. Cinnamaldehyde treatment had a positive effect on oxidative stress, inflammation and apoptosis levels and alleviated nephrotoxicity. This study demonstrated that Cinnamaldehyde has ameliorative activity on Doxorubicin-induced nephrotoxicity.
