Development of photosystem II based bionanoenergetic and biomemory applications

Abstract (EN)

In this study, firstly, isolation of the photosystem II (PS II) complex from spinach by using chemical degradation method has been provided and cryogel based solid support material has been developed for purification. For this purpose, cryogel columns have been prepared by polymerization of hydroxy ethylmethacrylate (HEMA) monomer including phenylalanine and cross-linker N,N' methylenebisacrylamide (MBA) in the first step of the study. Then, characterization of the synthesized cryogel solid support has been performed. In addition, the determination of molecular weight and protein content of the purified PS II by using prepared column has been also carried out by SDS-PAGE and Bradford method, respectively. In the second step of the study, bioenergetics have been synthesized by using purified FS II. For this purpose, photosensitive micro-emulsion polymerization method has been applied. The prepared bionanoenergetics have been characterized by fluorescence spectroscopy, circular dichroism (CD) spectroscopy and zetameter. In the last step of the study, new approaches have been developed to nano PS II complex investigating biomemory and redox properties of the bionanoenergetics by cyclic voltammetry and chronoamperometry.
