Abstract (EN)
There are some basic problems brought about by systemic drug use. The main ones are the inability to ensure effective biodistribution of the drug in the body, the lack of affinity of the drug for the diseased area, the necessity of high systemic dose to reach sufficient local concentration, side effects and toxicity due to high drug doses. Until the drug reaches the site of action, it may lose its effectiveness by having to overcome many healthy organs, tissues, cells and barriers. Drug targeting, that is, predominant drug accumulation at the target site, can solve most of these problems. In this thesis, it is aimed to design, characterize and perform drug targeting applications of a nanocarrier to ensure that the relevant drug is directed to a specific target region using an external magnetic field at an effective dose. Doxorubicin Hydrochloride (DOX), which is widely used in cancer treatment, was chosen as a model drug. Three different magnetic materials MCapIII, PEG@MCapIII and MCapIV were synthesized as DOX carriers. While MCapIV appeared to exhibit a pH-sensitive release profile, the pH difference for MCapIII and PEG@MCapIII had no significant effect on drug release behavior. It has been determined that PEG@MCapIII has a more controlled release compared to MCapIII and MCapIV and its burst release effect is reduced with polyethyleneglycol coating.
