Assesment of macroprolactin measurement in patients with hiperprolactinemia

Abstract (EN)
The aim of this study is to evaluate the idiopathic hyperprolactinaemic patients serum, who applied to endocrinology polyclinic and to measure the macroprolactin with PEG precipitation method in this patients.The study include 38 women and 5 men at total 43 patients.PRL level measured with ECLİA method.The PEG 6000 at % 25 was used with two different materials as distile water and phosphate buffer.After precipitation with PEG the PRL is measured in the supernatant.The recovery below the % 40 reffered as macroprolactinaemic.İn 43 patient, the macroprolactin of the six patients were positive.The total PRL levels were below the 100 ng/ml in the macroprolactin positive patients.The strong positive corelation was found between the monomeric PRL levels which measured with two different PEG precipitation methods.To confirm the accuracy of the prl measurement in control samples intraassay CV?s % were control 1; % 1 and control 2; % 1.3.Another hand at day to day control CV?s were, control 1; % 3.4 and control 2; % 2.5.In hyperprolactinaemic patients the macroprolactinaemia should be come to mind as an important reasen.In PEG precipitation method both the phosphate and distile water can be used and each labrotary should own strategy to determine the macroprolactin.
