Abstract (EN)
Objective: The effects of the L-Theanine (LTA) on the hepatic microcirculation during hepatic ischemia-reperfusion injury (IRI) has not been investigated yet. The aim of this study was to investigate the influence of LT on the hepatic IRI in rats. Materials and methods: We investigated the effect of LTA on liver IRI by applying 70% one hour ischemia and 4 hour reperfusion in the liver in Karadeniz Technical University Medical Faculty Surgical Research Center. The study was carried out on four groups. The groups are Sham+Salin, Sham+LTA, IR+Salin, IR+LTA. The groups are Sham+Salin, Sham+LTA, IR+Salin, IR+LTA. While the sham groups were given saline and LTA without IR, the IR groups were given saline and LTA after IR. LTA was given 3 days before the experiment and 150mg/kg LTA was given on the day of the experiment before ischemia and before reperfusion. Results: Ischemia-reperfusion injury LTA use decreases the serum activity of serum alanine aminotransferase (ALT), lactate dehydrogenase (LDH), and tumor necrosis factor alpha (TNF-α). Brings tissue inducible nitric oxide synthase (iNOS), tissue myeloperoxidase (MPO) and tissue malondialdehyde (MDA) levels to normal levels after ischemia. In addition, oxidized glutathione (GSSG) caused a significant increase in tissue activity of the H score for hypoxia-inducible factor 1-α (HIF-1α). It provides an increase in the tissue activity of reduced glutathione (GSH) in the liver, and by providing a significant decrease in the Perfuze Vessel Density (PVD) values that increase after ischemia, it both increased tissue blood supply and showed positive effects on antioxidant mechanisms. Conclusion: In this study, LTA administration prevents hepatic IRI. This effect has been demonstrated by reducing lipid peroxidation, tissue iNOS level and TNF concentration, as well as decreasing HIF 1-α staining level by increasing tissue blood circulation and restoring cellular antioxidant level. LTA shows these effects with its antioxidant and anti-inflammatory effects
