INVESTIGATION OF TOXIC EFFECTS OF QUERCETIN ANDEPIGALLOCATECHIN GALLATEON C6 GLIOBLASTOMA MULTIFORME CELLS AND EFFECTSOF CALCIUM FOR THESE MECHANISMSABSTRACTIn this study, mechanisms of effects of quercetin and epigallocatechin gallateon C6 glioblastoma multiforme cells and role of calcium for this mechanismswere searched.As testing methods MTT, Neutral Red, Sulforodamin-B, Acridine Orangewere used. Calcium phosphate (in homeopathic manner) and thapsigarginewere used for understanding of role of calcium for cytotoxic effects ofquercetin and epigallocatechin gallate. By unique application of quercetin andepigallocatechin gallate gave rise to dose and time dependent cytotoxic andapoptotic effects on C6 cells. While, combinations of quercetin with calciumphosphate brought about mitotic cell death, combinations of epigallocatechingallate with thapsigargine caused to caspase-independent cell death.Combination of epigallocatechin gallate with calcium phosphate could notalter apoptotic mechanisms of epigallocatechin. However, combination ofepigallocatechin gallate with thapsigargine gave rise to caspase-independentcell death like that of quercetin.As a conclusion, in this study, it was shown that cells can induce to differentcell death from apoptosis by perturbation of intracellular calcium balance.Key Words: quercetin, epigallocatechin gallat, thapsigargine, calciumphosphate, glioma, cell death.
