Investigation of chronic apelin treatment effect on liver antioxidant enzymes (CAT, SOD, GSH-Px)

Abstract (EN)

Apelin is a versatile peptide, secreting from adipose tissue and has a variety of physiological functions. Apelin affects many biological systems especially immune, cardiovasculer and neuroendocrine system. The effect of this peptide on some antioxidant enzymes in cardiomyocytes was investigated and found to increase the activity of catalase.In this study, the effect of apelin-13 on antioxidant system activity is investigated in the liver tissue, which is depot location for detoxification, synthesis and digestion of various substances. In this study 32 male Sprague-Dawley rats were divided into 4 groups. 1. group control (1 ml/kg/day saline solution), 2. group 30 µg/kg/day, group 3. 100 µg/kg/day and 4. group of 300 µg/kg/day and a single intraperitoneal dose of apelin 13 was injected for 15 days. In liver tissues taken the fifteenth day, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities and gene expression of these enzymes were measured. AP 30 was found lower in the SOD activities (p<0.05), AP 30 and AP 100 was found lower in the CAT activities, AP 100 and AP 300 was found higher in the GSH-Px activities than the control group (p<0.05). While any change wasn?t observed in the measurement of SOD and CAT gene expressions, the GSH-Px expressions of AP 300 group was found higher than the control group (p<0.05). The concentration of MDA in the AP 100 and AP 300 groups were found lower than the control group (p<0.05).As a result, it was suggested that apelin suppresed basal oxidative process by increasing GSH-Px activity and gene expression in liver tissue.
