Abstract (EN)

CD70 is known as tumor necrosis factor super family (TNFSF) molecule and ligand of another TNFSF costimulatory molecule of CD27. Immune cells are the cells that CD70 is expressed transiently, whereas CD70 expression has not been found on normal tissue cells. Although, several types of cancer showed CD70 positivity correlated with patient outcome, non-small cell lung cancer (NSCLC) requires further investigation. This study was designed to evaluate expression status of CD70, CD27, FOXP3 and CD3 in tumor microenvironment (TME) of NSCLC tissues, and the potential usage of soluble CD27 (sCD27) levels in serum as a diagnostic biomarker for cancer diagnosis. Protein expressions in TME was analyzed for 32 NSCLC tissues by Western Blot. sCD27 levels in serum of NSCLC patients was detected with ELISA. CD70 expression was found in 15.63% of overall samples. CD27 expression was detected in 75%, FOXP3 in 43.75%, CD3 in 87.5% in samples. sCD27 serum level was found significantly higher (p<0.0001) for the patient group with very high sensitivity (90%) suggesting that sCD27 has a potential usage as a diagnostic biomarker for NSCLC for detection of the disease. Only few NSCLC cases showed CD70 expression in TME. A high percentage of positivity in overall patient samples for CD27, FOXP3 and CD3 was not corelated with CD70 expression. Also, high level of sCD27 gave a significant result for CD70 positivity (p=0.007) suggesting CD70-CD27 interaction may cause CD27 cleavage and relase to serum in CD70+ tumors in NSCLC.
