Determination of lacosamide from pharmaceutical preparations using diode array detector coupled liquid chromatography

Abstract (EN)
Lacosamide is a new generation antiepileptic drug used to control seizures in patients with epilepsy. It helps to reduce the intensity and frequency of epileptic seizures by regulating the electrical activity of nerve cells in the brain. In this study, a new High-Performance Liquid Chromatography (HPLC) method was developed for the determination of Lacosamide. During the study, an Agilent Zorbax C18 (3.00×10 mm) column with a particle size of 3.5 µm was used. The mobile phase consisted of 50mM KH2PO4 and MeOH, and the method was operated at a flow rate of 1 ml/min. The column temperature was set at 30°C, and the injection volume was 3 µL. Detection of the analytes was performed using a diode array detector at a wavelength of 210 nm. The method was validated according to the ICH guidelines. Weighted regression calculations were performed, and a linearity graph was plotted for Lacosamide in the concentration range of 0.1 µg.mL-1 to 50 µg.mL-1 . The values for the limit of detection and limit of quantitation were calculated as 0.0622 µg.mL-1 and 0.1866 µg.mL-1 , respectively.
