Abstract (EN)
According to previous studies, it is shown that the human mesenchymal stem cells (hMSCs) have pluripotent properties. hMSCs can differentiate into cardiomyocytes, osteoblasts, chondroblasts, adipocytes, endothelial, hepatocyte and neuron cells, when hMSCs are exposed to certain physical and chemical stimulants. It is hoped that the differentiation of hMSCs into certain cell types in certain in vitro conditions can provide treatment for certain diseases and pathologic situation. The suitable cell culture conditions which are needed for differentiation of hMSCs into osteoblast, chondroblasts and adipocyte cells are known. Moreover, hMSCs can differentiate into different types of cells under mechanical stress (mechanical stretching, pressure, shear stress) as a physical stimulus. It was found that the tension on the cytoskeleton and/or in the junctions between the cells caused hMSCs to choose the certain cell types differentiation way. Our aim in this study is to investigate the gene expression profiles playing role in the differentiation of hMSCs with Tubacin under the mechanical stretching. 2µM Tubacin concentration produces maximum acetlyalted alpha tubulin at the end of 6 hours and this effect returns into basal level within 6 hours. It is founded that 2 ?M Tubacin does not affect adipogenic and osteogenic differentiation of hMSCs without mechanical stress for both short (6 hours) and long (1 day and 21 days) duration period. Both 6 hours 5 % mechanical strain with 0.25 Hz and 2 ?M Tubacinin cause decrease the adipogenic differentiation potential of hMSC by oil red o staining. Tubacin neither affects ostegenic and adiopgenic differentiation of hMSCs nor regulates these differentiations. In this study, it is shown that both PPARG and TFA2 which are known as the adipogenic differentiation markers, were significanlty decreased at mRNA level with both mechanical stress and Tubacin by using Real time PCR. Moreover, according to microarray analysis results mechanical stress changed mostly gene expression profile under tubacin treatment.As a result, mechanical stretching inhibits the adipogenic differentiation potential of hMSC with Tubacin.
