The effects of Metamizole, Diclofenac Sodium and Paracetamol on rats that have which experimental organophosphate poisoning

Abstract (EN)

Inhibition of AChE is primarily kept responsible for poisoning caused by Organophosphate (OP) compounds. In recent studies, it has been stated that OP poisoning linked to oxidative stress and inflammatory cascade in the mechanism of toxicity are important. Therefore, it has been claimed that antioxidant and anti-inflammatory agents reduce toxicity of OP poisoning.In this study, it is aimed to introduce the effects that Paracetamol and Diclofenac Na, Metamizole poisoning OP on the inflammatory system and oxidative stress by the oxidant-antioxidants and biochemical parameters. In this way, it will be evaluated that both whether or not they are reducing the damage connected to poisoning OP inflammatory system and oxidative stress that may develop over the organ and when usage indications within OP poisoning occur while clinical practice which analgesics within the context of the experiment may be safely preferred.Rats randomly divided into five groups icludind 8 rats in each group. Sham group rats were included in the experiment without any substance. By giving IP route Fenthion (0.2 g/kg) to group II rats OP poisoning has been created. After one hour a single dose of saline IP (given equal to drugs volume) were performed. Rats in this group were treated with no treatment. Group III rats was given IP route Fenthion (0.2 g/kg), by this way OP poisoning has been created and after an hour a single dose of IP Metamizole (40 mg/kg) was given. Group IV rats IP route Fenthion (0.2 g/kg) was given. After an hour OP poisoning has been created then a single dose of Paracetamol IP route (10 mg/kg) was given. Group V rats IP route Fenthion (0.2 g/kg) was given. OP poisoning has been created by this way and an hour later a single dose of IP Diclofenac Na (5 mg/kg) was given. In the 24th hour of the study, immediately after sacrificing the rats from whom blood samples were taken for serological analysis tissue samples from their heart, liver, lung, kidney, brain and pancreas were taken.After OP poisoning in rats, while the MDA, GSH, ceruloplasmin, SOD, GPx, CAT and TNF-? levels were statistically and significantly increased, OP Ascorbic Acid, Carotene and Retinol levels were statistically reduced in a meaningful way. After OP poisoning, the increase in IL-10 levels did not reveal statistically significant.Metamizol has been identified as the most effective drug statistically and significantly reducing the levels of MDA, SOD and increasing the levels of Retinol. Reducing the levels of Diclofenac Na; Ceruloplasmin, CAT and TNF-? and increasing the IL-10 levels statistically and significantly, Ascorbic Acid has been identified as the most effective drug. On the other way, Paracetamol did not show an effect which is more effective than other drugs on any parameters. Among three drugs a statistically significant effect on GSH and Carotene was not seen.Malondialdehid and GSH tissue level evaluation results show that the application of the treatment is done usually by blocking the effects of their increase in MDA is emerging. However, this effect can be variable depending on the organ is striking. Metamizol, in MDA-mediated therapy, has shown protective effect in kidney, lung and brain tissues which is statistically significant, while the GSH-mediated treatment only on the brain tissue showed a statistically significant protective effect. Paracetamol has shown a statistically significant and protective effect in MDA-mediated treatment of lung, and brain tissue, while it has not shown any protective effect in the treatment of GSH-mediated on any tissues. Diclofenac Na, caused a statistically significant protective effect in MDA-mediated treatment of liver, lung and brain tissues, while in GSH-mediated therapy it has shown statistically significant protective effect on the kidney and lung tissue.Consequently; Metamizole is more efficient in oxidative stress sourced damages, Diclofenac Na is more efficient in inflammatory cascade sourced damages. In the treatment of OP poisoning, Metamizole should be preferred especially for kidney, lung and brain effected patients, while Diclofenac Na should be preferred for liver, kidney and lung effected patients. However, this result is the first study on this issue so further studies with additional support is required.
