Analysis of MİR27A RS895819 mutation in morbid obesity

Abstract (EN)
Obesity is becoming a public health problem that has become increasingly common all over the world. Genetic factors associated with obesity are frequently investigated to improve the treatment of obesity. In this context, the relationships between miRNAs involved in the regulation of gene expression as well as mutations in structural genes and obesity are frequently discussed. The aim of this study was to evaluate the relationship between miR27a rs895819 mutation and morbid obesity. In this study, 57 patients diagnosed as morbidly obese and 55 healthy controls were examined. The mean age of the patient and control groups were 38.91±12.03 and 25.76±4.23 years, respectively. The mean body mass index of the patient group was determined as 40 kg/m2. DNA isolation was performed in blood samples of patients and controls and miR27a rs895819 region was examined by polymerase chain reaction method. The results were evaluated with Chi-square and Hardy-Weinberg equilibrium. As a result of the study, no significant correlation was found between miR27a A>G mutation and morbid obesity in terms of allele and genotype frequency distribution (p> 0.05).In conclusion, it has been found that the mutation of miR27a rs895819, which is the negative regulator of adipogenic pathways, is not associated with obesity formation processes in a limited Turkish population. The results obtained should be confirmed in larger and different populations in order to be certain. Keywords: Obesity, morbidity, body mass index, miR27a.
