Abstract (EN)

The growth parameters of shoot tips, internodal segments and leaf explants excised from an aromatic plant Origanum minutiflorum O. Schwarz et P. H. Davis belonging to Lamiaceae family, have been exclusively evaluated by using different growth media, and the and analysis of phenolic consitituents of the plantlets herewith were also investigated. Germination procedures were applied both in the light and the dark by using Murashige ve Skoog (MS, ½ MS, ¼ MS) and Gamborg?s (B5, ½ B5, ¼ B5) basal media. The best suitable medium was determined for the germination, shoot formation and development and callus formation, individually. The micropropagation potential of this plant was investigated by using Murashige ve Skoog (MS; 1962) basal medium supplemented with various plant growth regulators (PGR), such as 6-benzlyamino purine (6-BA) , thidiazuron (TDZ) and kinetin. The best germination was observed on MS medium in the presence of light. The highest callus in light medium yiels was observed on the same medium supplemented with 6-BA 2 mg/L and IBA 2 mg/L. The best shoot formation was obtained from the shoot tip explants being cultured on MS basal medium supplemented with 6-BA 1 mg/L + IBA 0,5 mg/L. The extracts obtained from the plantlets grown eight weeks in vitro were also analysed by HPLC for their three major phenolic constituents.
