Research of Carvacrols Effects on Lower Extremity Ischemia/Reperfusion Injury on Rats Aim: Acute extremity ischemia is a clinical problem that causes high morbidity and mortality. Studies show that carvacrol has various biological and pharmacological properties, including antioxidant, antibacterial, antifungal, anticancer, anti-inflammatory, hepatoprotective, spasmolytic and vasorelaxant. In our study, it was aimed to investigate the effects of carvacrol on Total Oxidant Status (TOS), Total Antioxidant Status (TAS), and Thiol Disulfide Homeostasis (TDH) in femoral ischemia-reperfusion injury. Materials and Methods: 36 Spraque Dawley male rats were used in the study and 6 groups (Control, Ischemia-reperfusion, Carvacrol, DMSO, Heparin and Heparin+carvacrol) were formed, with 6 animals in each group. Except for the control group, the animals were anesthetized with 90/10 mg/kg ketamine/xylazine IM application, and then they were treated with rubber tape for 60 minutes. left femoral ischemia was created. Waited 60 min for reperfusion. During the reperfusion phase, the groups given carvacrol were given 0.01mL (5mg/kg) ip of carvacrol, and the groups given carvacrol heparin were given 0.1mg/;kg enoxaparin sc. TAS, TOS, Thiol Disulfite Levels were evaluated by ELISA in 5 ml intracardiac blood taken after reperfusion, and FIR was evaluated in terms of cell apoptosis, irregularity of muscle fibers, hemorrhage and inflammatory cell increase by hematoxylin-eosin coloring by excluding the lower extremity from the caput femoris level. Findings: In our study, biochemically and histopathologically, TAS, TOS, Thiol Disulfide Levels were increased, degeneration, cell separation, inflammation and congestion findings were found to be significant, and our data were consistent with the literature findings. The findings of disorganization, hemorrhage and necrosis were found to be statistically insignificant. Results: In our study, it was concluded that carvacrol has positive effects on ischemia-reperfusion injury. More experimental and clinical studies are needed to reveal the full implications of our findings and to apply them in practice.
