The effects of clotrimazole on experimental acute necrotizing pancreatitis in rats

Abstract (EN)

Aim: The aim of this study was to investigate the influence of clotrimazole on the systemic effects of pancreatitis and effects on pancreatic tissue and microcirculation on acute necrotizing pancreatitis(ANP) induced by cerulein and glycodeoxycholic acid in rats.Material and Methods: Sixty four male Sprague-Dawley rats weighing 300-350 grams were used. The rats were divided into five experimental groups. Group 1 (sham+salin, n=10), Group 2 (sham+KLT, n=16), Group 3 (sham+ polyethylene glicol, n=6), Group 4 (ANP+salin, n=16), Group 5 (ANP+KLT, n=16). The acute pancreatitis was induced by an intravenous infusion of cerulein and a standars infusion of glycodeoxycholic acid into the biliarypancreatic duct. Six hours after the induction of pancreatitis clotrimazole was given to treatment group. At the end of the experimental time pancreatic blood flow was measured with orthogonal polarisation spectrometry and then blood samples were obtained from the sacrified rats for blood gases analysis, IL-6 and biochemical parameters. The bronchoalveolar lavage(BAL) was performed and samples were obtained for BAL LDH. The left lung and pancreas were harvested for the measurements of myleperoxidase (MPO) and malonildialdehide (MDA) levels and histopathological evaluations. The histopathological evaluations were done by pathology specialist who does not informed about evaluated groups. The edema, inflammation and necrosis of pancreas were assessed.Results: Pancreatic tissue microcirculation levels were higher in the clotrimazole group (p<0.005). In the histopatholgoic evaluation clotrimazole decreased pancreatic edema and necrosis (p<0.005). Serun BUN, alanin aminotransferase and calcium levels and mean blood pressure and urine volume was improved with clotrimazole treatment (p<0.005). The use of clotrimazole decreased MPO and MDA levels in the lung and pancreatic tissue (p<0.005).Conclusion: This study demonstrates that clotrimazole has a beneficial effect on the cure of ANP in rats.
