The effects of intravenous pgi2(iloprost)on ischemia reperfusion injury in skeletal muscles on rats

Abstract (EN)

Aim: The aim of this study is to investigate the effects of prostoglandin I2 derivate which called iloprost, in the model of lower extremity skeletal muscle on rats in ischemia reperfusion injury.Method: Twenty-four Wistar Albo rats were randomized into six groups (n=4).Laparotomy was performed in all groups under general anesthesia. Only laparotomy applied in Group S (Sham). Ischemia reperfusion group (Group I/R)underwent ischemia and reperfusion performed by clamping and declamping of theinfrarenal abdominal aorta for 120 minutes, respectively. Group iloprost (Group I) received intravenous infusion of iloprost 10 mcg/kg respectively, without ischemiaand reperfusion. Group I/R/I received intravenous infusion of iloprost 10mcg/kg respectively immediately after 2 hours period of ischemia. At the end of reperfusion period, skeletal muscle samples of lower extremity were taken from all groups for biochemical and histopathologic examinations.Result: Tissue levels of endotelyal nitrikoksit (eNOS) were significantly higher inI/R groups than SHAM and Group I. In Group I/R/I, the levels of eNOS significantly high but, lower than the I/R group.The heat shock protein 60 (HSP 60) levels on Group I/R were higher than the other groups. But I/R/I groups results were found as same with the other twogroups.Malondialdehit (MDA) enzymatic activities were found significantly high in I/R group. On the other hand in; I/R/I group, MDA activities higher than SHAM and I group but lower than The I/R group.When superoksit dismutaz (SOD) enzymatic activities were examinated, the results were found meaningfully different in all groups. SOD enzymatic activitieswere found in I/R group, meaningfully lower than the other there groups. Also in group I/R/I, the SOD activity were higher than group I/R. But, İ/R/İ group SOD enzymatic activities were forunda higher than in İ/R group but, lower than inSHAM and iloprost group.Conclusion: These results indicate that iloprost have protective effects on ischemia reperfusion injury in skeletal muscle.
