The effects of antiepileptic drugs on experimental Acute Necrotizing Pancreatitis in rats

Abstract (EN)
1. INTRODUCTION AND AIM: Pancreatic necrosis is one of most important parameter in acute pancreatitis that predicts surveillance. Preventing cellular necrosis to control clinical progress and during acute pancreatitis the mechanisms leading apoptosis are important in prognosis. Even though fatal hemorrhagic pancreatitis cases have been noticed, it is not clear that V is a risk factor for pancreatitis. Relationship between pancreatitis and use of T did not noticed in literature. Importance of inflammation and apoptosis in pancreatitis physiopathology is clear. and In this study, we aim to investigate whether V has negative and T has positive effect on pancreatitis and if it exists which mechanisms in histology and pathology and effects on pancreatic necrosis and apoptosis.MATERIAL AND METHODS: The study was designed with 6 groups and in sham groups 6 rats, in pancreatitis groups 7 rats has been used. The acute pancreatitis was induced by an intravenous infusion of cerulein and a standart infusion of glycodeoxycholic acid into the biliary-pancreatic duct. Topiramate (80 mg/kg i.p.) to the sham+T and ANP+T group and valproic acid (300 mg/kg i.p.)to the sham+V and ANP+V were given simultaneously 6th and 18th hours in three days before the procedure including the day that pancreatitis made. In the fourth day cardiac and biochemical parameters were measured. The bronchoalveolar lavage (BAL) of the right lung was performed and LDH in BAL was measured. At the end of the experiment MPO and MDA activity in pancreas and also histological examination of pancreas were evaluated.RESULTS: When edema, necrosis and inflammation of ANP group compared with other groups, inflammation ratio of Group 1, Group 2, Group 3 and necrosis ratio of Group 1 and Group 3 were signicifantly increased (P<0.0033). Only inflammation in group 5 was signicifantly increased when compared Group 1, Group 2 and Group 3 (P<0.0033). Edema, necrosis and inflammation in were signicifantly increased between group 6 and sham groups (P<0,0033). Edema, necrosis and inflammation were signicifantly increased between pancreatic groups and sham groups . (P<0.05).CONCLUSION: The study shows topiramate has an improved outcome and valproic acid has no beneficial effect on acut pancreatitis regarding histopathological scores with statistically significance.Key Words: Acute necrotizing pancreatitis, apoptosis, Topiramate, valproic acid
