Abstract (EN)
In this thesis, the effects of biotic and abiotic elicitors (AgNP and MeJA) on photosynthetic pigment content, lipid peroxidation degree, proline content, total phenolic and flavonoid substance contents and total antioxidant activity in Salvia nemorosa L. plant exposed to NaCl stress factor were evaluated. For this purpose, after the seeds sown in pots reached a certain size, they were watered with ¼ Hoagland nutrient solution containing 100 mM NaCl for 10 days. At the end of this period, the pots were divided into five groups as “Control, 100 mM NaCI, 100 mM NaCI + 50 µM MeJA, 100 mM NaCI + 10 µM AgNP and 100 mM NaCI + 50 µM MeJA + 10 µM AgNP” and the plants were harvested after the 10th day. When the results were compared with the control group in terms of photosynthetic pigment contents (chlorophyll-a, chlorophyll-b and total carotenoid), the highest values were obtained from NaCI + AgNP + MeJA extract, in which both elicitors were applied together. The increase in the amount of MDA, which is a measure of lipid peroxidation, has been in the form of “NaCI > NaCI + AgNP > NaCI + MeJA > NaCI + AgNP + MeJA > Control” from high to low. Proline content increased in all treatments compared to the control group and the lowest amount was in NaCI + AgNP + MeJA application with 3.954 mmol/g TA, the highest value (5,874 mmol/g TA) was in 100 mM NaCI application containing only salt stress factor. In terms of total phenolic and flavonoid substance amounts, the highest content was NaCI + AgNP + MeJA; the lowest content was observed in the NaCI application. In addition, when the applications are examined in terms of total antioxidant activity evaluated by three different methods; In the DPPH method, the activity value of NaCI + MeJA application at low concentrations (100 to 150 µg/mL) was found to be higher than the positive control, ascorbic acid. The DPPH radical scavenging activity of the extracts from the applications in which elicitors were used together and separately at 250 µg/mL concentration was higher than all positive controls (AA, BHT, BHA). In ABTS method, while all applications at low concentrations generally show higher activity than positive control BHT; ethanol extracts of plants in which elicitors were applied together and separately at concentrations of 60 to 80 µg/mL showed higher activity than positive controls. In the CUPRAC method, the radical scavenging activity of the extracts, especially at 150 and 200 µg/mL concentrations of the applications in which two elicitors were used together, was found to be higher than BHA. Elisitor applications generally increased the radical scavenging activity in all three total antioxidant methods compared to the group in which salt was applied alone. Therefore, in terms of the parameters we use, the positive results of the application of biotic and abiotic elicitor together led to the conclusion that they can be effective in eliminating the negative effects of stress conditions.
