Abstract (EN)

In this study, prokaryotic diversity of Van Lake was investigated. Samples were taken from central, Tatvan, Ahlat and Edremit coasts from Van Lake. Isolation was performed on %2 MGM, %3 MGM, %5 MGM, %10 MGM, %12 MGM, Alkaline Nurient Agar, Horikoshi I and II Medium, Starch-Casein Agar, Actinomycetes Isolation Agar and Glycerol Yeast Extract Agar. 16S PCR was carried out for pure to colonies on media and amplified ribosomal DNA restriction analysis (ARDRA) was applied the 16S PCR products. The nucleotid sequences of the purificated PCR products were sequenced and 40 different strains in 124 isolates for Van Lake were identified.16S rRNA gene sequences were compared with reference sequences at NCBI using BLAST. According to the BLAST results, Halomonas, Alkalimonas, Marinobacter, Vibrio, Rhodococcus in Bacteria domain were identified as dominant species. Denaturating gradient gel electrophoresis (DGGE) was performed for studying population dynamics. As result of this gel in sequence analysis Citrobacter sp. was identified. Flourocens in situ hybridization (FISH) was used for water samples. The community was dominated by Bacteria and some water samples showed positive results only with bacterial fluorescent hybridization probes. Prokaryotic diversity of Van Lake was characterized and existence of some strains were showed in Turkey. In addition detailed study about alkaliphiles was showed first time in our country.
