Effect of folic acid on mesenchymal stem cell differentiation in wound healing model

Abstract (EN)

Mesenchymal stem cells (MSC) are adult type stem cell where the heterogeneous subset of stromal stem cells that can be isolated from adult tissues. MSC can be a differentiator for both mesodermal cells and other embryonic lines. Due to their origin as stromal, they carry the supporting cell feature and are resistant cells with high potency in different areas. Studies show that the effects of transplanted MSC are mediated through differentiation properties or by the release of molecules that regulate immune cells, restoring tissue function. After in vivo applications, it has been shown that MSC induce peripheral tolerance and migrate to damaged cells and prevent the release of damaged area pro-inflammatory cytokines and promote the survival of damaged cells. Folic acid, another important factor in tissue regeneration and repair, has been questioned in the literature due to its role in the synthesis of building block molecules and its effects on differentiation. In this study, the effect of folic acid on mesenchymal stem cell differentiation was investigated in the wound healing model. In the wound healing model generated by the HUVEC cell line, tissue regeneration was assessed using folic acid and its inhibitors, MSCs secretory factors. The wound healing rate was measured by a real-time live cell imaging system and cell proliferation functions were measured by real-time cell proliferation. Increasing effect of folic acid on cell proliferation. MSH epidemic factors potentiate this increase during the first six hours. In this study, endothelial damage-enhancing capacities of factors that mesenchymal stem cells secreted co-culture endothelial with folic acid were investigated in an in vitro wound healing model. It has been shown that MSCs contribute not only to support cell functions but also to the effect of differentiation on the secretory functions in tissue regeneration and healing processes, and this contribution potentiates the effect of folic acid on tissue regeneration.
