Investigation of the effect of silver nanoparticles with primula vulgaris extract on MCF-7 cells by applying the green synthesis method

Abstract (EN)

In this study, silver nanoparticles (AgNP) were synthesized by green synthesis method using dried root, leaf and flower extract of Primula vulgaris plant. The cytotoxic effect of the obtained silver nanoparticles loaded with Primula vulgaris extract (PVLAgNP) on the MCF-7 cell line was evaluated. Visible Spectrophotometry (U-Vis), X-Ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), Scanning Electron Microscopy (SEM) and Zeta Potential measurement techniques were used in the synthesis and characterization processes of PVLAgNPs. The maximum absorption wavelength for AgNP solutions synthesized from root, flower and leaf was found to be 440 nm. It was determined that PVLAgNPs are crystalline structures according to XRD spectra. Zeta potential values of PVLAgNPs were found to be in the range of -14 to -28 mV on average. SEM analysis showed that synthesized PVLAgNPs were predominantly spherical in shape and particle size ranged from 22 nm to 52 nm. For the cytotoxicity activity of PVLAgNPs against MCF-7, iCELLigence (real time cell analysis system) was used. Inhibitory concentrations (IC50) of PVLAgNPs were found to be 30.37, 36.74 and 57.64 µg / mL for leaf, root and flower at 24 hours of incubation, respectively. When the results of this study were evaluated, it was determined that PVLAgNP has a significant cytotoxic effect on MCF-7 cancer cell lines.
