Abstract (EN)
With biological varieties found in nature, glucose has been found to be one of the most critical for unquestionable life. In this study; a conjugated liposomal structure targeting qualitative analysis of glucose has been developed based on 10,12-Pentacosadiynoic acid (PCDA) which forms a conjugated polydiacetylenic system exhibiting unique chromatic properties when polymerized. PCDA-NHS, a derivative of PCDA with a long aliphatic core and carbonyl and hydroxyl groups; was synthesized by NHS/EDC method. Characterization of PCDA-NHS was done by FT-IR, 13C NMR and 1H NMR analyzes. PCDA-NHS liposome was synthesized by thin-film hydration and sonication using PCDA-NHS, 2 methacryloyloxyethyl phosphorylcholine (MPC), and PCDA; Scanning Electron Microscopy (SEM), size and zeta potential analyzes were performed for characterization. The liposome was approximately 212.8 nm in size. The carbohydrate binding protein Concanavalin, with a molecular weight of 25.5 kDa and a size of approximately 462.6 nm, was conjugated to the synthesized liposome by incubation technique. After conjugation, the liposome dimensions were measured to be approximately 791.1 nm and the percentage of colorimetric response was calculated by glucose detection with these protein-conjugated liposomes. Also, the UV absorption analysis was performed. As a result, this liposomal structure is thought to have an important role in glucose detection for the future.
