Investigation and epidemiological typing of mycobacterium bovis in cattle with suspected tuberculosis in Adiyaman province

Abstract (EN)

Aim: The causative agent of bovine tuberculosis, M. bovis, threatens human health due to its zoonotic character. In this study, it was aimed to isolate M. bovis from animals slaughtered for meat production by point surveillance method in one year period in Adıyaman province and to determine profiles by spoligotyping. Materialandmethod: In the study, 44 tissue samples taken from cattle carcasses with typical granulomatous lesions after slaughtering between 10.10.2017 and 31.12.2018 in Adıyaman were examined. These samples were DHK (Decontamination Homogenization Concentration) treated with 4% NaOH-NALC method. DNA isolation was done using the Mickle DNA isolation device (Mickle Tissue Disintegrator). Microscopic examination was performed with EZN staining before and after DHK treatment. Conventional culture (Löwenstein Jensen with and without pyruvate) was inoculated on medium and evaluated by In-House PCR method.Colonies collected from the cultures were typed epidomiologically using the spoligotyping method. Results:The positivity rate of 44 tissue samples examined was microscopy 34.09% (15/44) before the DHK procedure, microscopy 40.90% (18/44 ) after the DHK procedure, culture 54.54% (24/44) in Löwenstein Jensen medium with pyruvate, and culture in Löwenstein Jensen medium without pyruvate 68.18% (30 / 44 ) and PCR tests were 90.90% (40/44). As a result of spoligotyping, 4 different epidemiological types were found (SB1197, SB1197, SB2508, SB2513). Conclusion: In the study we conducted with the point surveillance method, it was determined that tuberculosis was common in Adıyaman and that the tissue samples taken from cattle were M. bovis in 4 different epidemiological types.
