Abstract (EN)
In this thesis study, the protective effects of 3-benzoyl-7-hydroxy coumarin against oxidative stress induced by AlCl3 in rats were investigated. Four groups were formed in present study. These groups are Control group (K), Coumarin group (C), Aluminum group (A) and Coumarin + Aluminum (C+A) group. Coumarin as 10 mg/kg and AlCl3 as 8.3 mg/kg were applied every other day. MDA, reduced GSH, AChE, GST, Ces, some minerals and fatty acids were determined in the liver and brain tissues of rats. When the biochemical parameters of the liver tissue were examined, the levels of Al, Fe, Cu and MDA in group A increased whereas GSH, Ces, GST and Mn levels decreased. The levels of MDA, Fe, Al and Cu in the C+A group decreased compared to the A group. On the other hand, GSH and GST levels in C+A group increased compared to A group. When the biochemical parameters of the brain tissue were examined, Al and MDA levels of group A increased compared to group K. MDA and Al levels in C+A group decreased compared to A group. GSH, Ces, AChE and GST levels in A group decreased compared to the K group. Ces and AChE enzyme activity levels in C+A group increased compared to A group. When the fatty acid levels of brain tissue were examined, it was observed that 16:1, 20:4n6 and 22:6n3 fatty acid levels in group A decreased compared to the other groups. In addition, PUFA level of group A was found to be significantly decreased. In the C+A group, it was observed that these fatty acids increased with the effect of coumarin. When the fatty acid levels of liver tissue were examined, it was observed that the ΣSFA level of group A increased relative to the group K, while the level of ΣUSFA decreased relatively. In the C+A group, it was observed that USFA and PUFA levels increased with the effect of coumarin compared to the A group. As a result, we think that 3-benzoyl-7-hydroxy coumarin compound has positive effects as antioxidant against oxidative stress caused by AlCl3.
