Abstract (EN)

SUMMARY In this study, from mud and water of Ayder (Rize) hot spring, thermophilic bacteria isolation were carried out, and then, species of isolates were tried tobe determined. Meanwhile, the number of thermophilic bacteria in the samples from hot spring was determined. The number of bacteria in the main hot spring samples of the thermal was defined by the membran filtration method. In addition, the samples obtained by filtration and the enrichment cultures from the hot spring's water and mud were grown on nutrient agar medium and the colonies were examined under binocular microscope. Five different isolates were subjected to the further tests. Morphological, physiological, biochemical, some genetical and chemotaxonomical features of the isolates were examined. As conclusion, the number of Bacteria in the hot spring was found as 13 bacteria/ml. Since all isolates are Gram variable, aerobic, motile and spore forming bacteria, they are placed into genus Bacillus. Isolates have distinct features from the present species in the references. For the exact determination of the species, the results should be supported by the 16S rRNA gene sequence and %G+C content. Key words: Ayder Hot Spring, Thermophilic Bacterium, Bacillus sp. VII
