Abstract (EN)

The Gram-positive model prokaryote Bacillus subtilis is a non-pathogenic organism that is known for producing a wide variety of secondary metabolites with pharmacological and antimicrobial activities. The smallest dipeptide antibiotic bacilysin is among the many secondary metabolites that are produced by B. subtilis, and it is composed of L-alanine and L-anticapsin. As shown earlier by our research group, the synthesis of bacilysin is regulated by bacA operon under the control of quorum sensing and sporulation pathways. Additionally, our findings suggested that bacilysin acts as a pleiotropic molecule and alters the cellular physiology of B. subtilis. In the present study, the transcriptomic profiles of mutant OGU1 and its parental strain PY79 were investigated by RNA-Seq and validated with RT-qPCR analyses. While a total of 2369 significantly differentially expressed transcripts were identified by RNA-Seq, only 223 differentially expressed genes (DEGs) were overexpressed in mutant OGU1. The results of previous comparative proteomics studies were integrated with the current transcriptomic (RNA-Seq) analysis, and the results are compared and discussed with respect to their cellular functions. Overall, this study confirms and expands our earlier studies with a fresh perspective on the pleiotropic changes that emerge from the loss of bacilysin biosynthetic function.
