Determining the potential of some drugs to become cholinesterase inhibitors

Abstract (EN)

Acetylcholine (ACh) is the main neurotransmitter in the body that functions in both the peripheral and central nervous systems. Acetylcholinesterase (AChE, EC 3.1.1.7) is an enzyme from the hydrolase group that hydrolyzes the neurotransmitter ACh. Butyrylcholinesterase (BChE, EC 3.1.1.8) is a nonspecific cholinesterase enzyme. Cholinesterase inhibitors inhibit the enzymes that break down ACh. The main use of cholinesterase inhibitors is to treat dementia in Alzheimer's patients. ACh level in the brain decreases in Alzheimer's disease. Cholinesterase inhibitors play a role in curing the disease by balancing this level. In this thesis, it was tried to determine the inhibition potential of some drug active ingredients (Etofenamate, brimonidine, bimatoprost, tetrahydrozoline, trimethoprim and dexketoprofen) on AChE and BChE enzymes. For this purpose, docking and IC50 values were calculated for these drugs which show inhibition effect on enzyme activities.
