Abstract (EN)

SUMMARY In this study, it was aimed propagation of Betula medwediewii and Zelkova carpinifolia subsp. Yomraensis which have been difficult obtaining of the material, had few amount of individual and endemic species' by using tissue culture. It was studied on bud explants of Betula medwediwii taken south and north side of 6 different ortets in growing season. 40 samples were obtained for every ortet and they were cultured into the WPM (Woody Plant Medium) included 4 mg/1 BAP. 2, 6, and 10 weeks later from the explants were cultured, effects of the different clones and direction of taken exsplants from ortet were researched on the survival rate of the explants, callus and shoot formation. In the end of 6 weeks, formed long shoots were placed into WPM rooting media included 2 mg/1 NAA. And the shoots were rooted 4 weeks later. Bud explants taken from the young ortet during the dormancy season were cultured cultured into the WPM media included different BAP + NAA concentrations. The bud explants belonged Zelkova carpinifolia subsp. Yomraensis were placed into WPM and MS (Murashige & Skoog) media included Zeatin, BAP, IAA, and IBA concentrations and combinations. Callus formation was realised but shoot formation was not realised for Zelkova carpinifolia subsp. Yomraensis. Key Words: B. medwediewii, Z. carpinifolia subsp.
