Phylogenetic analysis of some taxa balonging to lamiaceae family in Bitlis province with RAPD-PCR technique

In this study, the relationship between 54 taxa of 21 genera beloning to Lamiaceae family which grow naturally in Bitlis province have been demonstrated. Samples collected from the province of Bitlis were defined morphologically and preserved in Bitlis Eren University Herbarium. Genetic similarities between taxa were determined by RAPD (Randomly Amplified Polymorphic DNA) PCR technique. In the study, 50 different RAPD primers were tested and among them 9 primers showing high polymorphism were evaluated. According to the results obtained, Ajuga L. and Teucrium L. strains belonging to the subfamily Ajugoideae showed parallelism to the morphological system. Ajugoideae subfamily made a cluster with taxa that belong to Scutellarioideae subfamily. The genera that we studied were belonging to the subfamily of Lamioideae, namely Phlomis L., Lamium L., Ballota L., Stachys L. and Sideritis L. genera were supported the morphological systematics, while Marrubium L. genera seperated from the group. It was observed that taxa belonging to Nepata L., Lallemantia Fisch. & C.A. Mey, Melissa L., Prunella L., Origanum L., Satureja L., Clinopodium L., Cyclotrichium (Boiss.) Manden. & Scheng., Mentha L. and Salvia L. genera from Nepeteoideae subfamily supported morphological system, but Ziziphora clinopodioides Lam. taxa showed diference. On the other hand, Marrubium parviflorum Fisch. & C.A. Mey. subsp. parviflorum and Marrubium astracanicum Jacq. taxa have showed affinity of 0.900 ratio according to similarity matrix. Although Marrubium members belong to Lamioideae subfamily, they created unaccompanied group and stayed away from the other subfamily members. According to similarity matrix, the most similarity were found between Teucrium chamaedrys L. subsp. sinuatum Celak. Rech.f. and Teucrium polium L. subsp. polium with 0,908 ratio, between Phlomis lanceolata Boiss. & Hohen. and Phlomis kurdica Rech. Fil. with 0,908 ratio, between Origanum acutidens (Hand.-Mazz.) Ietswaart and Origanum vulgare L. subsp. gracile (K.Koch) Ietswwith 0,923 ratio, between Clinopodium vulgare L. subsp. arundanum (Boiss.) Nyman and Clinopodium graveolens subsp. rotundifolium (Pers.) Govaerts 0,955 ratio, between Salvia verticillata L. subsp. verticillata and Salvia verticillata subsp. amasiaca (Freyn & Bornm.) Bornm. with 0,934 ratio, between Salvia verticillata subsp. amasiaca ( Freyn & Bornm.) Bornm. and Salvia candidissima Vahl. subsp. candidissima with 0,917 ratio, between Salvia candidissima Vahl. subsp. candidissima and Salvia limbata C. A. Meyer with 0,900 ratio.
