Observation of the effects of lysosyme and silver anode application in raw cow milk on biofilm formation of Pseudomonas aeruginosa

Abstract (EN)

In this study, antibiofilm effects on Pseudomonas aeruginosa were investigated by using Silver Anode Technique and Lysozyme enzyme in raw milk. In addition, the antibacterial effects of lysozyme and silver anode technique on raw milk were observed at 0, 24, 48 and 72 hours storage periods, and the effect of silver anode technique on milk parameters was determined. In the study, lysozyme inhibited biofilm formation of Pseudomonas aeruginosa, which is found in the natural flora of raw milk, by 47%. It prevented the biofilm formation of Pseudomonas aeruginosa ATCC 27853 strain inoculated into raw milk by 40%. Under the same conditions, the antibiofilm effect of the silver anode technique resulted in 62% for Pseudomonas aeruginosa found in the natural flora of milk and 58% for Pseudomonas aeruginosa ATCC 27853 inoculated raw milk. The highest antibiofilm effect was seen in the designs where lysozyme and silver anode technique were used together as synergists. While 75% biofilm was prevented in raw milk containing Pseudomonas aeruginosa in its natural flora, 78% biofilm reduction was observed in raw milk inoculated with Pseudomonas aeruginosa ATCC 27853. Lysozyme alone showed bacteriostatic activity against Pseudomonas aeruginosa ATCC 27853 and Pseudomonas aeruginosa during storage periods. The silver anode technique, on the other hand, provided a decrease of up to 1 log at the first application, and a decrease of 1.5-2 logs of bacteria, especially at the 72nd hour of storage. With the combination of silver anode technique and lysozyme enzyme in raw milk, up to 1 log of bacteria reduction was observed at the time of application. The reduction continued at 24 and 48 hours of storage. A strong bactericidal effect was seen, ultimately resulting in a reduction of 2-2.5 logs at 72 hours of storage. The results obtained from the study showed that Lysozyme and Silver Anode Techniques may have an important role in the fight against the waste and biofilm of this bacterium in the dairy sector.
