(ÇİRİŞ) Advisor: Assistant Professor Dr. Tuba AYDIN 2019, Page: 43 Jury: Prof. Dr. Mehmet Serdar GÜLTEKİN Assistant Professor Dr. Ahmet Gökhan AĞGÜL Assistant Professor Dr. Tuba AYDIN In this thesis, the inhibitory effect of methanol extract of Asphodelus aestivus Brot. (Çiriş) was investigated on h-CAI and h-CAII isoenzymes. One triglyceride, one long chain unsaturated fatty acid and two phytosterol were purified from extract by chromatographic methods. The chemical structures of the purified sterols were characterized as β-sitosterol and β-sitosterol-3-O-β-D-glycopyranoside by 1D and 2D 1H-NMR and 13C-NMR spectroscopic methods. IC50 values of methanol extract were calculated as 15.54 μg / mL (R2 = 0.9654) and 13.33 μg / mL (R2 = 0.9569) for h-CAI and h-CAII isoenzymes, respectively. According to these results, methanol extract of A. aestivus Brot. was evaluated as medium h-CAI and h-CAII inhibitor. In this study, the inhibitory effect of A. aestivus Brot. was revealed on h-CAI and h- CAII isoenzymes for the first time. Among the people of the plant is used especially against hemorrhoids.
