Abstract (EN)

The biological active values of bee products such as honey, pollen and propolis are due to the polyphenols contained in it. The highest polyphenol-containing product among the bee products is propolis. Propolis is a natural mixture formed by collecting the resins in flowers and buds by bees and processing them with some enzymes and plays a role in the defense of the hive. Raw propolis obtained by scraping from the hives cannot be consumed directly. It is consumed after processing in different formulations as ethanolic extracts. With this study, it was aimed to determine the optimum conditions for the preparation of ethanolic propolis extracts and determination of the phenolic profile. The ideal extraction solvent was searched using ethanol / water mixture in different percentages from 0% to 100%. Propolis samples were evaluated according to total polyphenol (TP), total flavonoid (TF), total antioxidant (TA) capacity and brix values. The phenolic composition of propolis extracts were analyzed with the 25 phenolic standards in the HPLC-PDA system, considering the validation parameters such as precision, accuracy, LOD, LOQ and % recovery. As a result, it was determined that the optimum ethanolic solvent percentage varies between 60% and 70%, the ultrasonic extraction process increases the solubility and the most ideal propolis-solvent ratio (g/v) varies between 1: 5 and 1:10. It was found that the amount of phenolic acid increased as the polarity of the solvent used increased, whereas the amount of flavonoids increased as the polarity decreased.
