Evaluation of kinetic effects of gliotoxin in different breast cancer cell lines

Abstract (EN)

In this study the effects of Gliotoxin on Luminal A breast cancer and triple negative breast cancer cells were explored. In our investigation MCF-7 and MDA-MB-231 cell lines were used as Luminal A breast cancer and triple negative breast cancer model respectively. Growth rate, cell index values obtained from xCELLigence Real-Time Cell Analysis (RTCA) System, mitotic index, labelling index and apoptotic index were evaluated in experiments. In experiments, 1.5625 µM, 3.125 µM and 6.25 µM Gliotoxin concentrations were applied for both cell lines, and the concentration of 1.5625 µM was determined as the IC50 dose. As a result of the application of 1.5625 µM Gliotoxin concentration, it is seen that apoptotic index values of both cell lines increase depending on time. Mitotic index values of both MCF-7 and MDA-MB-231 cell lines at a concentration of 1.5625 µM Gliotoxin decreased significantly from the 24th hour. A significant time-dependent decrease in% BrdU values of both MCF-7 and MDA-MB-231 cell lines was observed in the Gliotoxin concentration of 1,5625 µM. When the data obtained from all applications that examined, a significant decrease was observed in the apoptotic index values, while a significant decrease in the rate of proliferation, cell index and mitotic index was observed in both cell lines with time.
