Determination of anti-melanogenesis activity of Glycine max cell suspension extract on B16-F10 melanoma cells

Abstract (EN)

In this study, it was aimed to determine the effect of soybean cell culture extract containing isoflavonoids and other phenolic compounds on the process of melanogenesis. For this purpose, firstly in vitro seedlings from soybean seeds were obtained and then callus and cell suspension cultures were prepared from them for further analysis. The total phenolic content in the extract of soybean cell culture was determined by Folin Ciocalteu method and isoflavonoids and the other phenolics were determined chromatographically by LC-MS/MS analysis. In vitro cytotoxicity of soybean cell culture extract was evaluated on mouse melanoma cells (B16-F10) and human normal fibroblast cells (CRL-2076). It was found that the soybean cell culture extract significantly (p<0,001) decreased melanin content and supressed tyrosinase activity on melanoma cells in a dose dependent manner. It was revealed that at both gene and protein levels soybean cell culture extract suppresses the expression of TYR, TYRP1 and TYRP2 enzymes by suppressing the expression of the MITF transcription factor. The results indicate that soybean cell culture extract has the potential to be an effective skin whitening agent.
