Determination of quercetin in jujube plants by HPLC-DAD

Abstract (EN)

In this study, quercetin was directly determined in jujube plants growing in the province of Kırklareli using high sensitivity HPLC-DAD without precipitation. During the method development phase, HPLC-DAD was used to optimize parameters such injection volume, wavelength, flow rate, and column temperature. With an ideal wavelength of 255 nm, quercetin was shown to have linearity within the linear range of 0.1–25 ppm, and the LOD LOQ values were found to be 0.02 ppm–0.07 ppm, respectively. The percentage relative standard deviation for intra-day and inter-day repeatability was as low as 0.1% and as high as 4.8%, respectively. The created technique was successfully used on jujube samples that were extracted using the proper technique. In addition, standard quercetin solution was added to the jujube samples for the accuracy of the method and >95% recovery was achieved. Keywords: Jujube, Quercetin, HPLC-DAD
