Investigation the effects of cytokines of young and old mesenchymal stromal cells on cancer cell lines by small molecules

Nowadays, stromal cell therapies are among the most important treatment methods used experimentally in many fields of medicine or clinically in some fields. Although different treatment methods are used, there is not yet a method that can cure cancer. Brain cancer is one of the difficult types of cancer to treat, and new methods are being researched for this. It has been reported in many experimental studies that Mesenchymal Stromal Cell (MSC), which has therapeutic and protective properties, suppresses cancer cells in different cancer models and stops their proliferation. However, in recent studies using stromal cells for cancer treatment; it has been shown that stromal cells trigger cancer formation, enlarge the mass, help the formation of new vessels, and invasion of cancer. It is known that communication between MSCs and cancer cells is achieved with the help of cytokines released from MSCs. The presence of these cytokines secreted by MSC and their effect on cancerous cells reduce the treatment reliability of MSC. However, it is found that MSCs are divided into two subgroups, one prevents the division and proliferation of cancer cells, while the other helps cancer cells to proliferate. Quercetin has been shown to play a positive role in many biological activities such as inflammation, oxidative stress, cancer formation and progression, platelet aggregation, atherosclerosis, and aging. This study aims to stimulate the release of cytokines and chemokines that will suppress cancer cells by stimulation of MSCs with Quercetin. The results showed that the conditioned MSC medium led to apoptosis and stopped the proliferation of the cells, giving different results for each patient for 24 and 48 hours. Two different cell lines were used in the experiment with these cell lines A172 and U87.
