Abstract (EN)

Since the 1990s, many publications have been made about metabolites of blood, urine and other fluids.However, presaturation pulse techniques have been used in these publications to suppress the water signal. These techniques are complex as they use multiple pulse steps. In these techniques, high-field NMR does not allow direct measurement of T1 and T2 relaxations. It also wastes experimental time, as they use high scan counts. The aim of this study is to obtain the metabolite profile of diabetic blood and urine samples at 400 MHz without water suppression and using a single pulse. 29 diabetic and 28 healthy blood and urine samples were collected from the clinics of Diyarbakır Memorial hospital. Healthy samples were used for comparative purposes. The blood mixtures were prepared by adding 0,02 ml of blood to 0.98 mL of D2O and urine mixtures were prepared by adding 0,06 mL of urine to 0,94 ml of D2O. The metabolite spectrum of each mixture was obtained by 400 MHz NMR. Only a 90degree radio frequency pulse was used in the experiment. 16 scans were made for each measurement. NMR spectrum of diabetic and healthy blood samples are not well dissolved and do not differ from each other. On the other hand, both healthy and diabetic urine samples gave a highly soluble spectrum with 400 MHz NMR and without water suppression. The number of peaks and the general profile of diabetic samples were different than healthy samples.This thesis study has shown that urinary metabolites can be obtained without using pre-saturation pulse techniques for water suppression, as well as without ultra-high NMR devices. The method used is simple and novel, and is suitable for the measurement of H2O T1 and T2 relaxation times. It also saves experimental time since it uses a low number of scans (16).
